as to your question as to how much torque a human hand can generate, I assume a rubber gloved hand (this is a surgical tool is it not?) that may be wet. not sure how that will affect the number but I suspect it will push the applied torque higher due to the higher COF between the rubber glove and the knob and the rubber and the skin on the other side. I would guess a number in the range of 50-75 ft-lb would be a good ballpark. unless overdesign is a real issue (it isn't aircraft parts after all, but if it is a high volume part it still might be) using a number in the range of 100 ft-lbs should be fine. (of course I used to work with a guy that had such strong hands, he could probably still break it if he really tried, but are you shooting for 100% or is 95% of the population good enough?)
